Endo-Sight™ Anti-Human IgG, VHH( pH-Red630) is a pH-sensitive fluorescent probe engineered for the real-time monitoring of antibody internalization. It combines a recombinant nanobody (VHH) specific to mouse IgG with the pH-Red630 fluorophore, which fluoresces only in acidic environments.
This probe is non-fluorescent at neutral extracellular pH but becomes intensely active upon internalization into acidic endosomes and lysosomes. It is a superior tool for studying the endocytic pathway and evaluating the trafficking efficiency of antibody-drug conjugate (ADC) candidates.
The Deep Red 630 probe features pH-sensitive fluorescence with excitation/emission maxima at 610/640 nm. Its fluorescence intensity is inversely proportional to pH, exhibiting a dramatic increase in signal within the pH 4.5–9.0 range. This makes it an ideal sensor for the acidic environments of endocytic vesicles. While the probe remains non-fluorescent in neutral extracellular conditions, it elicits a high-intensity signal upon internalization into acidic endosomal and lysosomal compartments.
